Guy (played by John Cassavetes) has a physically fragile wife, Rosemary (played by Mia Farrow). Ever since they moved into their new apartment, Guy has been getting along famously with a neighborly elderly couple. However, Rosemary does not particularly like the couple, and she is further unsettled by a series of strange occurrences. Rosemary sees a woman committing suicide, and under the increasingly bizarre atmosphere of her life, she begins to have chilling dreams. In her nightmares, a hairy monster assaults her, and shortly after waking from this nightmare, she learns she is pregnant. The elderly neighbor comes to care for Rosemary and her unborn baby; however, Rosemary gradually suspects that all the odd happenings stem from this seemingly kind and gentle neighbor, and even her husband! They are conspiring against Rosemary's baby. Is it just Rosemary's delusion, or is there something sinister going on? When she sees that the baby born has no pupils in its eyes, Rosemary realizes that the devil's presence is close at hand.
